Whole-blood samples were allowed to clot at 4℃ overnight and then centrifuged (1000×g, 4℃ ) for 20 min. Then, equal volumes of supernatant (serum) samples from each control group were mixed, while samples of each experimental group with equal volumes at the same time point were mixed, and then stored at -80℃. Following method of Li et al. [24], the complement C4 contents in fish serum were determined using kits from Hengyuan Ltd., Shanghai, China, according to the manufacturer’s instructions.
